What is Vyloy used for?
Vyloy (zolbetuximab-clzb) is commonly used to treat certain gastric (stomach) and esophagus cancers.
Vyloy may also be used for other conditions as determined by your healthcare provider.
How does Vyloy work (mechanism of action)?
Vyloy attaches itself to a specific protein called CLDN18.2 on the surface of cancer cells. After it attaches, it acts as a signal to your immune system to destroy the cancer cell. By killing the cancer cells, it can shrink tumors.
How is Vyloy supplied (dosage forms)?
Vyloy is available in an injectable form that is given by a healthcare provider.
How should I store Vyloy?
Vyloy is usually given by a healthcare provider in a hospital or clinic. You will not store it at home.
Side Effects
What are the most common side effects of Vyloy?
The most common side effects of Vyloy are listed below. Tell your healthcare provider if you have any of these side effects that bother you.
- Nausea and vomiting (see below)
- Tiredness
- Loss of appetite
- Diarrhea
- Nerve problems in your arms, hands, legs, or feet
- Stomach pain
- Constipation
- Weight loss
- Allergic reactions (see below)
- Fever
- Changes in blood cell levels
- Changes in kidney and liver function tests
- Low blood sugar
- Changes in electrolyte levels

What are the serious side effects of Vyloy?
While less common, the most serious side effects of Vyloy are described below, along with what to do if they happen.
Severe Allergic Reactions. Vyloy may cause allergic reactions, which can be serious. Get help right away if you have any of the following symptoms of a serious allergic reaction.
- Breathing problems or wheezing
- Racing heart
- Fever or general ill feeling
- Swollen lymph nodes
- Swelling of the face, lips, mouth, tongue, or throat
- Trouble swallowing or throat tightness
- Itching, skin rash, or pale red bumps on the skin called hives
- Nausea or vomiting
- Dizziness, feeling lightheaded, or fainting
- Stomach cramps
- Joint pain
Severe Nausea and Vomiting. Vyloy can cause severe nausea and vomiting and could lead to dehydration. Nausea and vomiting can happen at any time, but often happen during the first cycle (set) of Vyloy infusions. Your healthcare provider will give you certain medicines before each dose of Vyloy to help prevent nausea and vomiting. They may also give you medicine to have at home. Take these medicines exactly as your healthcare provider tells you. Tell your healthcare provider right away if you have severe nausea or vomiting.

Who should not use Vyloy?
Allergies to Ingredients. People who are allergic to any of the following should not use Vyloy.
- Vyloy
- Zolbetuximab-clzb
- Any of the ingredients in the specific product dispensed

What should I know about Vyloy before using it?
Vyloy is usually given by a healthcare provider in a hospital or clinic. Your healthcare provider will decide the dose that is right for you and how often you will receive Vyloy. They will also decide how many treatments you will need.
What should I tell my healthcare provider before using Vyloy?
Tell your healthcare provider about all of your health conditions and any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using. This will help them determine if Vyloy is right for you.
In particular, make sure that you discuss any of the following.
Nausea and Vomiting. Vyloy can cause serious nausea and vomiting, which can lead to other severe side effects. It is important to tell your healthcare provider if you have any nausea or vomiting before using Vyloy.
Pregnancy. It is not known if or how Vyloy could affect pregnancy or harm an unborn baby. Tell your healthcare provider if you are or plan to become pregnant.
Breastfeeding. It is not known if Vyloy passes into breast milk. You should not breastfeed while using Vyloy and for eight months after your last dose. Tell your healthcare provider if you are breastfeeding or plan to breastfeed.

Does Vyloy interact with foods or drinks?
There are no known interactions between Vyloy and foods or drinks.
It is unknown if drinking alcohol will affect Vyloy.

There are no known interactions between Vyloy and other medicines. Always tell your healthcare provider about any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using.
Overdose/Missed Dose
What should I do if I accidentally use too much Vyloy?
If you or someone else has used too much Vyloy, get medical help right away, call 911, or contact a Poison Control center at 800-222-1222.
What should I do if I miss a dose of Vyloy?
Vyloy is usually given by a healthcare provider in a hospital. Your healthcare provider will make sure you are receiving Vyloy as scheduled.
If you miss an appointment to get your Vyloy dose, call to make another appointment as soon as possible.
