What is Columvi used for?

Columvi (glofitamab-gxbm) is commonly used to treat certain types of diffuse large B-cell lymphoma or large B-cell lymphoma.

How does Columvi work (mechanism of action)?

Columvi is a type of medicine called immunotherapy. Immunotherapy works by turning on the body’s immune system so it can attack cancer cells. 

How is Columvi supplied (dosage forms)?

Columvi is available as an intravenous injection that is given by a healthcare provider.

How should I store Columvi?

Columvi is usually given by a healthcare provider in a hospital or clinic. You will not store it at home.

Side Effects

What are the most common side effects of Columvi?

The most common side effects of Columvi are listed below. Tell your healthcare provider if you have any of these side effects that bother you.

  • Cytokine release syndrome (see below)
  • Muscle and bone pain
  • Rash
  • Tiredness

What are the serious side effects of Columvi?

While less common, the most serious side effects of Columvi are described below, along with what to do if they happen.

Severe Allergic Reactions. Columvi may cause allergic reactions, which can be serious. Stop using Columvi and get help right away if you have any of the following symptoms of a serious allergic reaction.

  • Breathing problems or wheezing
  • Racing heart
  • Fever or general ill feeling
  • Swollen lymph nodes
  • Swelling of the face, lips, mouth, tongue, or throat
  • Trouble swallowing or throat tightness
  • Itching, skin rash, or pale red bumps on the skin called hives
  • Nausea or vomiting
  • Dizziness, feeling lightheaded, or fainting
  • Stomach cramps
  • Joint pain

Cytokine Release Syndrome (CRS). Columvi can cause a dangerous and sometimes life-threatening problem called cytokine release syndrome (CRS). CRS occurs when your immune system responds too much to medicines or infection. CRS may happen right away or several days after you get Columvi. You will receive a Patient Wallet Card that you should carry with you at all times. Get emergency help if you have any of the following symptoms of CRS.

  • Fever, chills, or shaking
  • Racing heart
  • Dizziness or feeling lightheaded
  • Trouble breathing or shortness of breath

Nervous System Problems. Columvi may cause nervous system problems that are serious and may lead to death. Call your healthcare provider right away if you have any of the following neurologic symptoms.

  • Headache
  • Confusion
  • Trouble paying attention or understanding things
  • Trouble speaking
  • Trouble remembering things
  • Numbness, tingling, or weakness in your hands or feet
  • Dizziness
  • Shaking

Serious Infections. Serious infections that can lead to death can happen if you are using Columvi. Keep all appointments to have your blood checked. Call your healthcare provider right away if you have any of the following symptoms of infection.

  • Fever
  • Chills
  • Feeling weak
  • Cough
  • Shortness of breath
  • Sore throat

Tumor Flare. Columvi can cause your tumor to grow or cause the symptoms of your cancer to get worse, which is called tumor flare. Call your healthcare provider right away if you have any of the following symptoms of tumor flare.

  • Lymph nodes that are sore or swollen
  • Pain or swelling at your tumor site
  • Chest pain
  • Cough
  • Trouble breathing

Warnings & Precautions

Who should not use Columvi?

Allergies to Ingredients. People who are allergic to any of the following should not take Columvi.

  • Columvi
  • Glofitamab
  • Any of the ingredients in the specific product dispensed

Your pharmacist can tell you all of the ingredients in Columvi. 

What should I know about Columvi before using it?

Columvi will be given to you by a healthcare provider through an intravenous (IV) line that goes into your vein. Your healthcare provider will tell you how many treatments you will need. 

Keep all appointments to check your blood work so that your healthcare provider can check for side effects.

Columvi may affect your alertness or coordination. Do not drive or do other activities that require alertness or coordination until you know how Columvi affects you.

What should I tell my healthcare provider before using Columvi?

Tell your healthcare provider about all of your health conditions and any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using. This will help them determine if Columvi is right for you.

In particular, make sure that you discuss any of the following.

Infections. Tell your healthcare provider if you have any signs or symptoms of an infection or a fever before receiving Columvi.

Kidney Problems. Before taking Columvi, tell your healthcare provider if you have any problems with your kidneys. 

Pregnancy. Columvi may cause harm to an unborn baby. You should have a pregnancy test before you start treatment. If you can become pregnant, use effective birth control (contraception) during treatment and for one month after your last dose of Columvi. If you become pregnant, contact your healthcare provider right away.

Breastfeeding. It is not known if Columvi passes into breast milk. Tell your healthcare provider if you are breastfeeding or plan to breastfeed. Do not breastfeed during treatment and for one month after your last dose of Columvi.

Interactions

Does Columvi interact with foods or drinks?

There are no known interactions between Columvi and foods or drinks.

It is unknown if drinking alcohol will affect Columvi. Columvi may cause nervous system problems, which may cause symptoms such as dizziness, confusion, or tiredness. The risk of these symptoms may be increased if you drink alcohol while taking Columvi.  

Does Columvi interact with other medicines (drug interactions)?

Always tell your healthcare provider about any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using.

Columvi may alter the blood levels of some other medicines. Tell your healthcare provider about all medicines that you take or have recently taken.

Overdose/Missed Dose

What should I do if I accidentally use too much Columvi?

If you or someone else has used too much Columvi, get medical help right away, call 911, or contact a Poison Control center at 800-222-1222.

What should I do if I miss a dose of Columvi?

Columvi is usually given by a healthcare provider in a hospital or clinic. Your healthcare provider will make sure you are receiving Columvi as scheduled. If you miss an appointment to get your Columvi dose, call to make another appointment as soon as possible.
