Augtyro (repotrectinib) is commonly used to treat certain kinds of the following cancers.

How does Augtyro work (mechanism of action)?
Augtyro is a type of medicine known as a tyrosine kinase inhibitor. It works by targeting specific proteins on cancer cells. By blocking the action of these proteins, Augtyro can cause cancer cells to die and can prevent cancer cells from growing or spreading.

How should I store Augtyro?
Augtyro should be stored at room temperature, between 68 F and 77 F (20 C and 25 C). It can be exposed to temperatures between 59 F and 86 F (15 C and 30 C) for shorter periods of time, such as when transporting it. Store in a cool, dry place.

What are the most common side effects of Augtyro?
The most common side effects of Augtyro are listed below. Tell your health care provider if you have any of these side effects that bother you.
- Tiredness
- Change in how things taste
- Numbness or tingling in your arms or legs
- Constipation

What are the serious side effects of Augtyro?
While less common, the most serious side effects of Augtyro are described below, along with what to do if they happen.
Severe Allergic Reactions. Augtyro may cause allergic reactions, which can be serious. Stop taking Augtyro and get help right away if you have any of the following symptoms of a serious allergic reaction.
- Breathing problems or wheezing
- Racing heart
- Fever or general ill feeling
- Swollen lymph nodes
- Swelling of the face, lips, mouth, tongue, or throat
- Trouble swallowing or throat tightness
- Itching, skin rash, or pale red bumps on the skin called hives
- Nausea or vomiting
- Dizziness, feeling lightheaded, or fainting
- Stomach cramps
- Joint pain
Central Nervous System (CNS) Problems. Augtyro can affect your central nervous system (CNS). Call your health care provider right away if you have the following symptoms of CNS problems.
- Dizziness
- Sleep problems
- Confusion
- Trouble with balance or coordination
- Seeing or hearing things that others do not see or hear, also called hallucinations
- Trouble with thinking, memory, and focus
- Changes in mood or behavior
Lung Inflammation. Augtyro can cause inflammation in your lungs, also called interstitial lung disease orpneumonitis. This can be serious or even life-threatening. Get emergency help if you have new or worsening breathing problems. You may also have a cough, wheezing, fever, or chest pain.
Liver Damage. Liver damage, also called hepatotoxicity, can happen when taking Augtyro. Keep all appointments to have your blood checked. Call your health care provider right away if you have any of the following symptoms of liver damage.
- Nausea or vomiting
- Stomach or belly pain
- Fever
- Weakness or unusual tiredness
- Itching
- Loss of appetite
- Light-colored poop
- Dark-colored urine (pee)
- Your skin or the whites of your eyes turning yellowish in color (also called jaundice)
Muscle Problems. Augtyro can cause muscle problems, including muscle pain, weakness, and tenderness, called myopathy. In some people, the muscle may break down (rhabdomyolysis), which can be serious and rarely lead to kidney damage and death. Get help right away if you have any of the following symptoms.
- Unexplained muscle pain, weakness, or tenderness
- Low energy levels or feeling easily tired, especially with activity
- Dark-colored urine (pee)
- Fever
- Muscle cramps
- Stomach pain
- Nausea or vomiting
High Levels of Uric Acid. Augtyro can cause high levels of uric acid (hyperuricemia) in your body. Tell your health care provider if you have had gout in the past or had high levels of uric acid. Attend all appointments for your provider to test your blood and urine so they can be sure that Augtyro is safe for you. Tell your health care provider right away if you have any of the following symptoms.
- Red, hot, tender, or swollen joints, especially in your big toe
- Stomach pain or pain in your sides
- Less urine (pee) than normal
- Nausea or vomiting
- Pink or brown pee or blood in your pee
Weak Bones. Augtyro can cause your bones to break (fracture) more easily. Tell your health care provider if you break a bone, have any pain, or have changes in your ability to walk or move.
Warnings & Precautions
Who should not use Augtyro?
Allergies to Ingredients. People who are allergic to any of the following should not take Augtyro.
- Repotrectinib
- Augtyro
- Any of the ingredients in the specific product dispensed

What should I know about Augtyro before using it?
Do not take Augtyro unless it has been prescribed to you by a health care provider. Take it as prescribed.
Do not share Augtyro with other people, even if they have the same condition as you. It may harm them.
Keep Augtyro out of the reach of children.
Swallow Augtyro whole. Do not crush, split, dissolve, or chew the capsules. If one of your Augtyro capsules is broken, cracked, or damaged, do not take the dose. Contact your pharmacist about getting a replacement capsule.
Augtyro can affect your alertness or coordination. Do not drive or do other activities that require alertness or coordination until you know how Augtyro affects you.
If you are able to get a partner pregnant, use condoms while taking Augtyro and for four months after your last dose.
What should I tell my health care provider before using Augtyro?
Tell your health care provider about all of your health conditions and any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using. This will help them determine if Augtyro is right for you.
In particular, make sure that you discuss any of the following.
Current and Past Health Conditions. Tell your health care provider if you have any of the following.
- Nervous system problems
- Previous bone fractures, weak bones, or osteoporosis
- Gout or high uric acid levels
- Lung or breathing problems
- Liver problems
- Kidney problems
Other Medicines and Supplements. Augtyro may interact with other medicines and supplements. Before taking Augtyro, tell your health care provider about any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using. See the Interactions section for more details.
Pregnancy. Augtyro may cause harm to an unborn baby. Your health care provider will give you a test to make sure you are not pregnant before you start taking Augtyro. If you can get pregnant, you should use nonhormonal birth control, such as condoms, while taking Augtyro and for two months after your last dose. Tell your health care provider if you are pregnant, think you may be pregnant, or plan to become pregnant.
Breastfeeding. It is not known if Augtyro passes into breast milk. You should not breastfeed while taking Augtyro and for ten days after your last dose. Tell your health care provider if you are breastfeeding or plan to breastfeed.

Interactions
Does Augtyro interact with foods or drinks?
Grapefruit interacts with Augtyro. Do not eat grapefruit or drink grapefruit juice while taking Augtyro. Grapefruit can interfere with your body’s ability to break down Augtyro, which can lead to high levels of the medicine and an increased risk of side effects.
It is unknown if drinking alcohol will affect Augtyro, but heavy drinking can slow your immune response and make it harder for your body to fight cancer.
Does Augtyro interact with other medicines (drug interactions)?
Always tell your health care provider about any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using.
In particular, make sure that you discuss if you are using any of the following before taking Augtyro.
- Itraconazole (Sporanox, Tolsura), which is a medicine used for fungal infections
- Rifampin (Rifadin), which is a medicine commonly used to treat an infection called tuberculosis
- St. John’s wort, which is a supplement that may be used for depression
- A birth control pill, patch, vaginal ring, implant, injection, or intrauterine device (IUD) that contains hormones
Many other medicines may alter the blood levels of Augtyro and vice versa. Tell your health care provider about all medicines that you take or have recently taken.

Overdose/Missed Dose
What should I do if I accidentally use too much Augtyro?
If you or someone else has used too much Augtyro, get medical help right away, call 911, or contact a Poison Control center at 800-222-1222.
What should I do if I miss a dose of Augtyro?
If you miss a dose, skip the missed dose and only take the next dose. Do not take double or extra doses.
If you vomit after taking your Augtyro dose, do not take another dose. Skip the dose and take your next dose at the usual time.
